The alleged father and child must be tested
For best results the child's mother is typically tested as well

The test is based upon the principles of inheritance. A child
gets one half of his/her genetic makeup from the mother and
the other half from the biological father.
The test analyzes the inheritance of genetic markers. The
genetic markers that the child shares with the mother are
first located. Then to determine paternity, the child's remaining
markers are compared to the alleged father. If the man is
indeed the father, all of the markers that did not match the
mother will match his. If all of the child's remaining markers
match the alleged father, evidence is provided that he is
the biological father of the child. If they do not match,
he is excluded as the biological father of the child.
When the mother is not tested, a larger batch of genetic markers
must be analyzed from the alleged father and child. If these
markers show that the man is contributing half of the genetic
makeup of the child, he is given evidence that he is the biological
father of the child. If no markers are found to match between
the alleged father and child, the man is excluded from being
the biological father of the child.

The PI is a measure of discrimination used in paternity analysis
and is a means for presenting the genetic odds in favor of
paternity given the genotypes for the mother, child and alleged
father.

The
routine use of blood samples is on the decline for paternity
testing. Buccal (oral mouth or cheek) swabs are rapidly becoming
the specimen of choice. The specimen is collected by gently
rubbing the cheeks inside the mouth with long swabs similar
to Q-tips. The buccal cells that come off in this process
do not require refrigeration, preservatives, or immediate
shipping to the laboratory. In addition to the easy collection,
the specimen is useful after years of storage. The DNA tests
utilized on buccal swab specimens are the same DNA test as
used on blood specimens.
Buccal
swab specimens are not affected by bacteria, toothpaste, chewing
tobacco or other tobacco products, lipstick, or nursing (mother's
milk). The testing is not affected by bacterial DNA as bacteria
do not contain the DNA sequences examined in the test. No
fasting is required prior to specimen collection.
Genetic
Assays has experienced no recollections of the buccal swab
specimen. The result in all cases reported to date are either
clear exclusions or probabilities of paternity greater than
99%.
